55. Identification of Mutations Responsible for Hereditary Neurodegenerative Disorders in Dogs

Description: The neuronal ceroid lipofuscinoses (NCLs) are a group of inherited diseases characterized by progressive degeneration of the brain, retina, and other parts of the nervous system. Affected dogs exhibit symptoms that include movement problems, visual impairment, seizures, loss of training, and personality changes. The disorders usually lead to premature death. NCLs have been reported in many breeds. To date, we have identified gene defects responsible for NCL in English Setters, American Bulldogs, and Dachshunds. We currently offer screening tests to identify the gene defect in these breeds. The gene defect that underlies NCL in Border Collies has also been identified. Using an approach similar to that used to identify the genetic defects in these breeds, we propose to identify the genetic defects responsible for NCL in Tibetan Terriers, Australian Shepherds, Miniature Schnauzers, Polish Lowland Sheepdogs and, possibly, other breeds if suitable samples can be obtained. These studies will utilize molecular genetic tools we developed in our previous studies of NCLs and other inherited diseases in dogs. The proposed studies will lead to the development of breed-specific DNA screening tests that will enable breeders to identify carrier and affected dogs prior to breeding and thus to eradicate NCL from affected breeds.
